Construction and Application of Quantitative Detection Method for Pathogenic Bacteria in Wheat Stem Base Rot
To further identify the incidence of wheat crown rot in Tianjin area and preform disease prevention and control,areal-time PCR for quantitative detection of Fusarium pseudograminis and Bipolaris sorkiniana was established.The specific primers Fpg-qrt-F/R and Bs-qrt-F/R amplified the fragment of 298 bp and 116 bp were designed based on the rDNA ITS sequence of F.pseudograminis and B.sorkiniana.A standard curve established using genome of pathogens,was used to investigate the content of F.pseudograminis and B.sorkinianain wheat rhizosphere soil and seed samples in Tianjin area.Using real-time PCR detection system,thegenomic con-tent of F.pseudograminis and B.sorkiniana in wheat rhizosphere soil was 0.343 8-77.686 7 pg·g-1 and 1.685 3-844.229 0 pg·g-1.In addition,the detection rate of F.pseudograminis in wheat seeds samples of 15different areas was 40%,with pathogen content range of 0.261 7-0.269 0 pg·g-1.And there was no B.sorkiniana dectected.The results indicated the incidence of wheat crown rot in Tianjin area and determined the minimum threshold of pathogen in soil,which provide an early warning of the occurrence of soil-borne dis-eases.
