白芸豆淀粉酶抑制剂提取工艺优化及与左旋肉碱复配物功效评价

为优化白芸豆淀粉酶抑制剂提取物(α-AI)的提取工艺,并探究白芸豆α-AI-左旋肉碱复配物的减质量效果及安全性,以白芸豆α-AI提取物活性为检测指标,以白芸豆粉碎粒度、浸提液pH、料液比、浸提温度、浸提时间为单因素,在单因素的基础上设计响应面优化白芸豆α-AI提取工艺;通过减肥功能评价方法、大鼠急性经口试验,验证复配物的减质量效果及安全性.结果表明:白芸豆提取物在浸提粉碎粒度为80目、浸提液pH值为6、温度为53℃、浸提时间为3.3 h、料液比为1∶7条件下提取工艺最佳;对照组与模型组体质量差异显著,且复配物的急性毒性试验未见异常.本研究可为白芸豆提取物的高效提取及减质量配方的开发提供新思路与数据支撑.
Optimization of Extraction Process for α-AI from White Kidney Beans and Evaluation of Its Efficacy in Combination with L-carnitine
To optimize the extraction process of white kidney bean amylase inhibitor extract(α-AI)and explore the weight loss effect and safety of white kidney bean α-AI-L-carnitine complex,this study used the activity of white kidney bean α-AI extract as the detection indicator,and used white kidney bean powder particle size,extraction solution pH,solid-liquid ratio,extraction temperature,and extraction time as single factors.Based on these single factors,a response surface methodology was designed to optimize the ex-traction process of white kidney bean α-AI.Verify the weight loss effect and safety of the compound through weight loss function e-valuation methods and acute oral tests in rats.The results showed that the optimal extraction process for white kidney bean extract was as follows:extraction powder with a particle size of 80 mesh,extraction solution pH=6,temperature of 53 ℃,extraction time of 3.3 hours,and solid-liquid ratio of 1∶7.There was a significant difference in body weight between the control group and the model group,and no abnormalities were observed in the acute toxicity test of the compound.This study can provide new ideas and data support for the efficient extraction of white kidney bean extract and the development of weight loss formulas.
