Research on Shoot Tip Culture and Rapid Propagation Technology of Niuwei Yam
In order to establish a virus-free culture and rapid propagation system for Niuwei yam,suitable culture media for shoot tip culture,stem segment propagation,and rooting culture were screened using tissue culture techniques,and the appropriate conditions of acclimatizating plantlets were studied.The results showed that the expansion rate and survival rate of shoot tips were higher in the medium MS+0.5 mg·L-16-BA+1.5 mg·L-1NAA.The optimal concentration of polyvinylpyrrolidone(PVP)for inhibiting shoot tip browning was 0.01%,with a browning rate of only 11.67%.The medium MS+1 mg·L-16-BA+0.3 mg·L-1NAA was better for adven-titious bud emergence from a stem segment with a node.In the rooting culture,the medium MS+0.5 mg·L-16BA+1 mg·L-1NAA was the most appropriate.The optimal acclimatization conditions for yam plantlets were mixed substrate(perlite:straw humus=1:1),temper-ature of 20℃,shading rate of 75%,and relative humidity of 80%.In summary,this study lays the foundation for virus-free culture and propagation of Niuwei yam,which is of great significance for the sustainable development of yam industry.
