白芍总苷对H2O2诱导下黑色素细胞的保护及作用机制

为了探究白芍总苷治疗白癜风的潜在作用机理,对人正常黑色素细胞PIG1进行体外培养,用不同浓度的白芍总苷对细胞进行预处理并施加H2O2刺激,用CCK-8法检测细胞活力,用流式细胞术检测细胞凋亡和活性氧的产生,用蛋白免疫印迹法检测细胞色素c、cleaved caspase-3、cleaved caspase-9含量以及MAPK家族成员p38及其磷酸化的表达。结果显示:①白芍总苷可有效抑制H2O2诱导的PIG1细胞毒性,且呈剂量依赖性;②白芍总苷减少了H2O2诱导的细胞活性氧的产生和细胞色素c的释放,下调了cleaved caspase-3和cleaved caspase-9的激活;③ 白芍总苷有效阻碍了H2O2诱导的p38 MAPK磷酸化。上述结果表明,白芍总苷可作为一种有效的抗氧化剂,保护黑色素细胞免受H2O2诱导的氧化应激损伤。
Protective effect of total glucosides of paeony on melanocytes induced by H2O2 and its mechanism
To explore the potential mechanism of total glucosides of paeony(TGP)in the treatment of vitiligo,human melanocytes PIG1 cells were cultured in vitro,which were pretreated with different concentrations of total glucosides of paeony and stimulat-ed by H2O2.CCK-8 assay was adopted to detect cell viability,flow cytometry was used to detect cell apoptosis and ROS produc-tion,and Western blotting was used to detect the expression of cytochrome c,cleaved caspase-3,cleaved caspase-9 protein,as well as MAPK family member p38 and its phosphorylation.The results were as follows:① TGP can effectively ameliorate the H2O2 induced oxidative stress damage in PIG1 cells in a dose-dependent manner;② TGP reduced cell ROS production and the release of cytochrome c,and downregulated activation of cleaved caspase-3 and cleaved caspase-9;③ The pretreatment with TGP hindered H2O2 induced p38 MAPK phosphorylation in a time-dependent manner.The above results demonstrate that TGP can be used as an effective antioxidant to protect melanocytes from H2O2 induced oxidative stress injury.
