Protective effect of total glucosides of paeony on melanocytes induced by H2O2 and its mechanism
To explore the potential mechanism of total glucosides of paeony(TGP)in the treatment of vitiligo,human melanocytes PIG1 cells were cultured in vitro,which were pretreated with different concentrations of total glucosides of paeony and stimulat-ed by H2O2.CCK-8 assay was adopted to detect cell viability,flow cytometry was used to detect cell apoptosis and ROS produc-tion,and Western blotting was used to detect the expression of cytochrome c,cleaved caspase-3,cleaved caspase-9 protein,as well as MAPK family member p38 and its phosphorylation.The results were as follows:① TGP can effectively ameliorate the H2O2 induced oxidative stress damage in PIG1 cells in a dose-dependent manner;② TGP reduced cell ROS production and the release of cytochrome c,and downregulated activation of cleaved caspase-3 and cleaved caspase-9;③ The pretreatment with TGP hindered H2O2 induced p38 MAPK phosphorylation in a time-dependent manner.The above results demonstrate that TGP can be used as an effective antioxidant to protect melanocytes from H2O2 induced oxidative stress injury.
