Forsythoside A inhibits the inflammatory damage effect of bEECs by negatively regulating the expression of HMGB1
Endometritis in dairy cows is one of the major diseases causing huge economic losses in the dairy industry.Recent studies have shown that forsythoside A,extracted from the traditional Chinese herb forsythia,has a wide range of biological activities.The aim of this study was to elucidate the inhibitory effect of forsythoside A on Escherichia coli endotoxin mediated inflammatory damage in bo-vine endometrial epithelial cells(bEECs)as well as the possible molecular mechanisms in vitro.The results showed that conotoxin sig-nificantly attenuated the damage effect of bEECs in a model of LPS-induced inflammatory injury in bEECs.Results of qPCR showed that forsythoside A significantly inhibited the release of pro-inflammatory cytokines(TNF-α,IL-6 and IL-1β)in a dose-dependent manner.Results from immunofluorescence and immunoblotting confirmed that coniferin significantly inhibited the expression of high mobility protein(HMGB1),which in turn blocked the activation of nuclear factor NF-κB.In conclusion,the findings suggest that forsythoside A protects against LPS-induced inflammatory injury in bEECs by negatively regulating HMGB1 expression,mediated by the NF-κB pathway.
