Construction of a prokaryotic expression vector and expression of the sheep pox virus 81 gene protein
To investigate the biological function of the sheep poxvirus 81 gene,a prokaryotic expression vector was constructed to expess its protein.Bioinformatics analysis of the 81 gene was conducted using online software,afollowed by gene amplification through PCR and the creation of a recombinant plasmid with the prokaryotic expression vector PET-42b.The protein was expressed in colon receptor cells.The study revealed that the 81 gene protein lacked a signal peptide and transmembrane region,contained 19 antigenic determinants,and was predicted to have an irregularly coiled,hydrophobic secondary and tertiary structure.PCR successfully amplified the 81 gene,and the PET-42b-81 prokaryotic vector was able to express the 18.35 kDa 81 gene protein.These results lay the groundwork for further exploration of the core genes of sheep poxviruses.
