溶磷菌的溶磷、分泌IAA及有机酸特性研究

对百脉根(Lotus corniculatus L.)根际不同部位溶磷菌进行分离,得到溶磷菌20株。通过溶磷圈法筛选出11株溶磷能力较强的菌株进行液体培养,利用钼蓝比色法测定其溶磷能力。结果表明,各菌株溶磷量在94.35~400.49μg mL-1之间,其中菌株LC15的溶磷量达到400.49μg mL-1。对菌株分泌IAA与有机酸的测定结果显示,菌株溶磷量与培养液有机酸总量、pH值之间不存在显著相关性。11株溶磷菌大部分具有分泌IAA的能力,其中菌株LC20分泌IAA量达到42.39μgmL-1,LC15分泌有机酸量较高,为30.67 mmol L-1,仅次于LC14(32.34 mmol L-1)。综合菌株的溶磷、分泌IAA和有机酸等特性,认为LC15、LC16、LC19、LC20和LC25等5个菌株具有较大的应用潜力。
Characters of Solubilizing Phosphate,Secreting IAA and Organic Acids of Phosphorus-Solubilizing Bacteria from Rhizosphere of Lotus corniculatus
Phosphate solubilization property of phosphobacteria strains,isolated from rhizosphere of Lotus corniculatus,was determined by using phosphate-solubilizing zone(PKO inorganic culture medium and Mengjinna organic culture medium) and phospho-molybdate blue color methods.Of the 20 isolated strains,11 strains possessed higher phosphate-dissolving capacity,with the range of dissolving phosphate from 94.35 to 400.49 μg mL-1,among which strain LC15 showed the highest phosphate solubilization capacity.The further experimental results showed that there was no significance relativity between phosphate solubilization capacity and the total quantity of organic acids in strain fermentation broth or the pH value,but most of the 11 strains had the ability to produce IAA,among them stain LC20 had the maximum IAA-producing capacity of 42.39 μg mL-1,and the capacity of secreting organic acids of strain LC15 was 30.67mmol L-1,just below that of the stain LC14,which was 32.34 mmol L-1.In terms of all the properties of dissolving phosphate and secreting IAA and organic acids,strains LC15,LC16,LC19,LC20 and LC25 could be used as prospective strains for Lotus corniculatus inoculant.
