赤芍根际耐盐碱促生菌分离鉴定及促生效果研究

为丰富赤芍根际促生菌(Plant growth promoting rhizobacteria,PGPR)菌种资源,本研究采用功能性培养基分离、筛选和鉴定赤芍根际促生菌,并通过种子萌发和幼苗生长实验评价其促生效果.研究以盐碱地赤芍根际土壤为材料,筛选出具有促生功能的耐盐碱菌株C14和C31.通过形态学和分子生物学鉴定,发现这两个菌株分别为嗜气芽孢杆菌(Bacillus aerophilus)和暹罗芽孢杆菌(Bacillus siamensis),且菌株C14在促生方面的效果明显优于菌株C31.种子萌发实验显示,在C14菌株(107 CFU·mL-1)的处理下浸种 48 h,赤芍种子的发芽率、发芽势、发芽指数和活力指数分别提高 29.5%、44.0%、30.6%和 55.5%.C14菌株处理后,赤芍根长、干重和鲜重在一定程度上均有所增加.研究结果为进一步探索耐盐碱促生菌的功能及盐碱地改良提供了后备菌种资源.
Isolation,identification and effect of saline-alkali tolerant growth-promoting bacteria in rhizosphere of Radix Paeoniae Rubra
To enrich the growth-promoting bacteria resources in the rhizosphere of Radix Paeoniae Rubra,functional medium was used to isolate,screen and identify rhizosphere growth-promoting bacteria,and their growth-promoting effects were evaluated by seed germination and seedling growth experiments.In this study,rhizosphere soil of Radix Paeoniae Rubra root in saline-alkali soil was used,and saline-alkali tolerant strains C14 and C31 with growth promotion function were screen out.The two strains were identified by morphology and molecular biology as Bacillus aerophilus and Bacillus siamensis,respectively.Strain C14 performed significantly better than strain C31 in promoting growth,and then C14 was selected for further study.The seed germination experiment showed that the germination rate,germination potential,germination index and vitality index of Radix Paeoniae Rubra seeds with the treatment of C14 strain(107 CFU·mL-1)for 48 h were increased by 29.5%,44.0%,30.6%and 55.5%,respectively.After treat-ment with C14 strain,root length,dry weight and fresh weight also increased to some extent.These results provide a reserve strain resource for further exploration of the function of saline-tolerant growth-promoting bacteria and improvement of saline-alkali soil.
