Cloning of large yellow croaker(Larimichthys crocea)FcRs gene and preliminary functional analysis in immune response
Fc receptors(FcRs)are a class of receptor that specifically binds to the Fc segment of immunoglobulin and mediates a variety of immune responses.In this study,six FcR genes were cloned from large yellow croaker(Larimichthys crocea),including LcFcγRⅠL,LcFcγRⅡ,LcFcγRⅡaL,LcFcγRⅢ,LcFcRL5 and LcFcRLA,and their open reading frame is 1 070,2 486,1 145,1 955,4 125 and 1 659 base pair,respectively.Each of LcFcRs contains a signal peptide and 2-8 Ig domains,and LcFcγRⅡ,LcFcγRⅢ and LcFcRL5 also contain a transmembrane region.Expression prolife analysis showed that LcFcγRⅠL,LcFcγRⅡ,LcFcγRⅡ aL and LcFcγRⅢ were mainly expressed in mucous tissues such as gill or intestine,while LcFcRL5 and LcFcRLA were mainly ex-pressed in immune tissues such as spleen and head kidney.Moreover,upon LPS the expression of these six FcRs genes in the spleen and head kidney was significantly up-regulated at different time points.After stimulation with poly(I:C)challenged,the six FcRs genes were significantly up-regulated in head kidney and spleen at different time points,whereas the expression of LcFcRL5 was significantly up-regulated in the spleen but significantly down-regulated in the head kidney.Results reveal that large yellow croaker FcRs may play different important roles in the immune response induced by LPS and poly(I:C).
