In Silico Cloning and Analysis of UDP-arabinopyranose Mutases Gene in Panax Ginseng
UDP-arabinopyranose mutase is a key enzyme in the process of arabinose synthe-sis,which plays an important role in plant morphogenesis and response to adversity stress.UDP-arabinopyranose mutase can interconvert UDP-Araf and UDP-Arap and participate in the biosynthesis of polysaccharides including arabinose.Using the amino acid sequence of the UDP-arabinopyranose mutase of Maize as a probe,the EST database of Panax ginseng was searched by silicon cloning to obtain a cDNA sequence with a high degree of homology,which was spliced and assembled using the DNAMAN software,and a cDNA sequence with a length of 1445 bp was finally obtained.Bioinfor-matics analysis of the UDP-arabinopyranose mutase gene of Panax ginseng was carried out using the relevant softwares.The results showed that the UDP-arabinopyranose mutase protein of Panax gin-seng is a stable hydrophilic protein,there are 33 phosphorylation modifications on the protein,no trans-membrane helical region,no signal peptide,and it is a non-secretory protein.The protein contains ir-regularly coiled,α-helix,extended chain,and β-folded secondary structures,of which the irregularly coiled structure is the most abundant.The amino acid sequence of the UDP-arabinopyranose mutase of Panax ginseng has a higher homology with Fenugreek and lower with Pomegranate.The analyzed re-sults can provide certain reference basis for the subsequent related studies on Panax ginseng.
