miR-431-5p通过MAPK/ERK通路促进食管鳞癌细胞增殖和迁移

为了检测miR-431-5p在食管鳞癌(Esophageal squamous cell carcinoma,ESCC)中的表达,探讨其功能,利用TCGA公共数据库分析miR-431-5p在食管癌中的表达,通过RT-qPCR检测miR-431-5p在组织和细胞中表达;再利用miR-431-5p模拟物和抑制剂,通过CCK-8 实验、细胞划痕和Transwell实验以及蛋白免疫印迹实验,检测miR-431-5p对ESCC细胞增殖、侵袭和迁移的影响以及分子作用机制.结果显示:miR-431-5p在食管癌中高表达;转染miR-431-5p模拟物后,KYSE30 细胞增殖率、迁移率和侵袭率分别增加了 24.19%,30.86%和 50.29%(P<0.05);而转染miR-431-5p抑制剂后,TE-11 细胞数量、迁移率分别降低了 21.43%和 9.52%(P<0.05);miR-431-5p模拟物增加KYSE30 细胞p-ERK表达量11.68%;miR-431-5p抑制剂降低TE-11 细胞p-ERK表达量 45.73%.研究结果表明:miR-431-5p通过MAPK/ERK通路促进ESCC细胞的增殖、侵袭和迁移.该研究为开发食管鳞癌治疗的有效靶点提供实验依据.
miR-431-5p Promotes the Proliferation and Migration of Esophageal Squamous Cell Carcinoma Cells via the MAPK/ERK Pathway
To detect miR-431-5p expression in ESCC and explore its function,we analyzed the TCGA public data-base for miR-431-5p expression in esophageal cancer.RT-qPCR was used to detect the expression of miR-431-5p in ESCC tissues and cells.CCK-8 assay,cell scratch and transwell migration assay and Western blotting were performed to detect the effect of miR-431-5p on the proliferation,migration and invasion of ESCC cells suffering the transfection of miR-431-5p mimics and inhibitors.Our results showed that miR-431-5p was highly expressed in esophageal cancer.Af-ter transfection of miR-431-5p mimics,the proliferation rate,migration rate and invasion rate of KYSE30 cells were in-creased by 24.19%,30.86%and 50.29%(P<0.05);while the number and migration rate of TE-11 cells were decreased by 21.43%and 9.52%(P<0.05)after transfection of miR-431-5p inhibitors;miR-431-5p mimics increased the expression of p-ERK in KYSE30 cells by 11.68%;miR-431-5p inhibitor decreased the expression of p-ERK in TE-11 cells by 45.73%.In summary,miR-431-5p promotes the proliferation,migration and invasion of ESCC cells through the MAPK/ERK pathway,providing experimental basis for the development of effective targets for the treatment of ESCC.
