Transcriptomic Analysis of Ferroptosis-Related Gene and Immune Infiltration in Multiple Sclerosis
Objective To investigate the immune infiltration and integrated network regulation of multiple sclerosis(MS)by transcriptomic analysis.Methods The differentially expressed ferroptosis-related genes of MS was identi-fied and the enrichment analysis was performed by GO and KEGG.LASSO and SVM-RFE algorithms were used to identify key ferroptosis-related genes in MS.ROC curve analysis was generated to test the predictive val-ue of the key ferroptosis-related genes in MS.MS mouse model was constructed to verify their expression by qRT-PCR.The CIBERSORT algorithm explored the correlation of key ferroptosis-related genes and im-mune cell infiltration.Online databases and Cytoscape were applied respectively to predict candidate small mole-cule compounds/drugs,transcription factors and miRNA.Results 39 ferroptosis-related genes of MS were se-lected and they were found associating with oxidative stress.BRD7,MTDH and SMAD7 were identified as the ferroptosis-related genes of MS.These three genes had high diagnostic efficacy and were all increased in the MS mouse model compared with in the control group.Besides,they were highly associated with immune infiltration.Conclusion BRD7,MTDH and SMAD7 were closely related with immune infiltration.
