Tangeretin regulates the proliferation and apoptosis of gastric cancer cells BGC-823 and SGC-7901 via PI3K/Akt/mTOR signaling pathway
Objective To investigate the effect of Tangeretin on autophagy of gastric cancer cells BGC-823 and SGC-7901 and its mechanism.Methods After treating the two kinds of cells with different concentrations of Tangeretin(7.5,15,30,60,120 μmol/L),the absorbance value was detected by CCK-8 method to calculate the cell prolifera-tion inhibition rate.Flow cytometry was used to detect cell apoptosis level;treated with Tangeretin at a concentration of 60 μmol/L to detect the migration ability of the two cells by cell scratching method.Western blotting method was used to detect autophagy-related protein p62,apoptosis protein Bax,Bcl-2 and PI3K/Akt/mTOR signaling pathway.The influ-ence of pathway related protein expression.Results Compared with the control group,the proliferation and migration ability of the Tangerine group was significantly decreased(P<0.01).The number of apoptosis increased,and the ex-pression of autophagy protein p62 increased in a concentration-dependent manner(P<0.05).The expression of apopto-sis protein Bax increased(P<0.05),Bcl-2 expression decreased(P<0.05),and the expression levels of phosphoryla-ted p-Akt and p-mTOR decreased significantly(P<0.05).Conclusion Tangeretin can regulate the proliferation,ap-optosis and migration of SGC-7901 and BGC-823 gastric cancer cells by inhibiting the PI3K/Akt/mTOR signaling path-way,and can inhibit autophagy.
