橘红素经PI3K/Akt/mTOR信号通路调控胃癌细胞BGC-823和SGC-7901的增殖和凋亡

摘要
目的 探讨橘红素对胃癌细胞 BGC-823 和 SGC-7901 的自噬作用及其机制.方法 不同浓度橘红素(7.5、15、30、60、120 μmol/L)处理两株细胞后,CCK-8 法检测其吸光度值,计算细胞增殖抑制率;流式细胞仪检测细胞凋亡水平;用 60 μmol/L 浓度的橘红素处理后通过细胞划痕法检测两种细胞迁移能力;Western blotting 法检测自噬相关蛋白 p62、凋亡蛋白 Bax、Bcl-2 及 PI3K/Akt/mTOR信号通路相关蛋白表达的影响.结果 与对照组相比,橘红素处理组增殖、迁移能力显著下降(P<0.01),细胞凋亡数增加,自噬蛋白 p62 的表达浓度依赖性升高(P<0.05),凋亡蛋白 Bax表达增加(P<0.05),Bcl-2 表达下降(P<0.05),磷酸化的 p-Akt和 p-mTOR的表达水平显著降低(P<0.05).结论 橘红素可通过抑制 PI3K/Akt/mTOR 信号通路调控 SGC-7901 和 BGC-823 细胞的增殖和凋亡,并可抑制其自噬.
Abstract
Objective To investigate the effect of Tangeretin on autophagy of gastric cancer cells BGC-823 and SGC-7901 and its mechanism.Methods After treating the two kinds of cells with different concentrations of Tangeretin(7.5,15,30,60,120 μmol/L),the absorbance value was detected by CCK-8 method to calculate the cell prolifera-tion inhibition rate.Flow cytometry was used to detect cell apoptosis level;treated with Tangeretin at a concentration of 60 μmol/L to detect the migration ability of the two cells by cell scratching method.Western blotting method was used to detect autophagy-related protein p62,apoptosis protein Bax,Bcl-2 and PI3K/Akt/mTOR signaling pathway.The influ-ence of pathway related protein expression.Results Compared with the control group,the proliferation and migration ability of the Tangerine group was significantly decreased(P<0.01).The number of apoptosis increased,and the ex-pression of autophagy protein p62 increased in a concentration-dependent manner(P<0.05).The expression of apopto-sis protein Bax increased(P<0.05),Bcl-2 expression decreased(P<0.05),and the expression levels of phosphoryla-ted p-Akt and p-mTOR decreased significantly(P<0.05).Conclusion Tangeretin can regulate the proliferation,ap-optosis and migration of SGC-7901 and BGC-823 gastric cancer cells by inhibiting the PI3K/Akt/mTOR signaling path-way,and can inhibit autophagy.
