Sophora japonica L.flower exosomes attenuate hypoxia-induced acute oxidative stress injury to the stomach and small intestine
Objective To analyze the role of Sophora japonica L.flower exosomes in hypoxia-induced acute oxidative stress injury to the stomach and small intestine. Methods Sophora japonica L.flower exosomes were extracted using ultracentrifugation followed by characterization through transmission electron microscopy and particle size measurement.Hypoxia-induced acute gastric and intestinal injury was triggered using placing mice in a ventilated hypoxic chamber sys-tem for 3 days.PBS or Sophora japonica L.flower exosomes were orally administered on day 1 under hypoxia.Mice were euthanized on day 3.Food consumption, body weight, stool score and stool blood score were recorded daily. The gastro-intestinal tract was isolated for HE staining. The gastric and small intestinal mucosa was collected for ROS detection. Results Sophora japonica L.flower exosomes treatment ameliorated acute gastric and small intestinal mucosal injury and downregulated ROS induced by hypoxia. Conclusion Oral administration of Sophora japonica L.flower exosomes effectively attenuated hypoxia-induced acute oxidative stress injury to the stomach and small intestine.
