摘要
目的 探讨UTP23在肝细胞癌中的预后价值及内在机制.方法 使用癌症基因组图谱(The Cancer Genome Atlas,TCGA)、基因表达综合数据库(Gene Expression Omnibus Database,GEO)和肿瘤免疫评估资源数据库(Tumor Immune Evaluation Resource,TIM ER)等公共数据库,分析肝细胞癌中UTP23的表达水平;采用Kaplan-Meier法评估UTP23的预后价值;使用Spearman法评估UTP23与肝细胞癌增殖、迁移相关因子表达的相关性.通过划痕实验和克隆形成实验分别研究敲低UTP23对HepG2细胞迁移和增殖的影响.结果 TCGA和GSE76427数据集中,肝细胞癌组织UTP23表达量相较于正常组织上调;TCGA中,UTP23高表达的患者总生存期(overall survival,OS)和无病生存期(disease free survival,DFS)较短(P=0.0056、0.011).UTP23与肿瘤增殖和迁移分子的表达呈正相关(P均＜0.05).细胞划痕实验显示UTP23敲低明显降低HepG2细胞的迁移能力;克隆形成实验显示,敲低UTP23明显降低HepG2细胞的克隆形成能力.结论 UTP23高表达与肝细胞癌发生发展以及患者不良预后相关.
Abstract
Objective To explore the prognostic value and intrinsic molecular mechanism of UTP23 in hepatocellu-lar carcinoma.Methods Public datasets such as The Cancer Genome Atlas(TCGA),Gene Expression Omnibus Data-base(GEO)and the Tumor Immune Evaluation Resource(TIMER)database were used to analyze the expression of UTP23 mRNA.Kaplan-Meier method was used to evaluate the prognostic value of UTP23.The Spearman method was used to evaluate the correlation between UTP23 and the expression of hepatocellular carcinoma proliferation-related fac-tors and tumor migration-related factors.The effects of UTP23 knockdown on HepG2 cell migration and proliferation were investigated by cell scratch test and plate cloning,respectively.Results In the TCGA database and GSE76427,UTP23 mRNA expression in hepatocellular carcinoma tissues was up-regulated compared with that in normal tissues.In TCGA,patients with high UTP23 expression had shorter overall survival(OS)and disease free survival(DFS),respec-tively(P=0.0056,0.011).In addition,UTP23 was positively correlated with the expression of cell proliferation and migration molecules(all P＜0.05).The cell scratch test showed that UTP23 knockdown significantly reduced the migra-tion ability of HepG2 cells.Plate cloning showed that the knockdown of UTP23 significantly decreased the clonogenic capacity of HepG2 cells.Conclusion The high expression of UTP23 is associated with the occurrence and development of hepatocellular carcinoma and the adverse prognosis of patients.
