The impacts of circMAN1A2 on the proliferation,invasion,and migration of gastric cancer cells by regulating the miR-212-3p/DDIT4 axis
Objective To investigate the impact of circMAN1A2 on the proliferation,invasion and migration of gas-tric cancer cells by targeting the miR-212-3p/damage-inducible transcript 4(DDIT4)axis.Methods The expression levels of circMAN1A2 and miR-212-3p in gastric cancer tissues and cells were detected by qRT-PCR.Luciferase assays were conducted to determine the targeting relationship between circMAN1A2 and miR-212-3p,as well as DDIT4 and miR-212-3p.Gastric cancer cells SGC-7901 and HGC-27 were transfected with si-NC,si-circMAN1A2,si-circMAN1A2+anti-NC,si-circMAN1A2+anti-miR-212-3p,miR-NC and miR-212-3p mimics,respectively,while non-transfected SGC-7901 and HGC-27 cells were used as controls.Cell proliferation was assessed using a plate cloning experiment,and the expression of proliferation,migration,and invasion-related proteins was measured by Western blotting.Results High expression of circMAN1A2 and low expression of miR-212-3p were observed in gastric cancer tissues and cells.Down-regulation of circMAN1A2 resulted in the up-regulation of miR-212-3p expression and the down-regulation of DDIT4 ex-pression,thereby inhibiting the proliferation,invasion,and migration abilities of gastric cancer cells.Inhibition of miR-212-3p expression partially reversed the inhibitory effect of circMAN1A2 expression on malignant proliferation of gastric cancer cells.Conclusion Inhibition of circMAN1A2 expression in gastric cancer cells can suppress the proliferation,mi-gration and invasion of gastric cancer cells.This effect is linked to the regulation of the miR-212-3p/DDIT4 signaling axis.
