Establishment of diabetic gastroparesis rat model by streptozotocin combined with high sugar and high fat diet and its evaluation
Objective To explore the method of establishing the animal model of diabetic gastroparesis(DGP).Methods Fifteen out of 25 normal SD rats were randomly selected for modeling.The rats were injected with 50 mg/kg 2%streptozotocin(STZ)solution once intraperitoneally to establish the diabetes model.After successful modeling,dia-betic rats were fed with high-sugar and high-fat diet for 6 weeks and were set as model group.Ten rats as control group were given a single intraperitoneal injection of 0.1 mol/L sodium citrate buffer(pH=4.5)and fed basic diet for 6 weeks.The general condition,body weight changes,random blood glucose changes,gastrointestinal dynamics indexes and morphological changes of HE-stained gastrointestinal sinus section were observed in the two groups of rats.Results Af-ter modeling,compared with the control group,rats in the model group had significantly lower body weight(P<0.01),significantly higher random blood glucose(P<0.01),lower gastric emptying rate(P<0.01)and lower propulsion rate of the small intestine(P<0.05);and disorganized arrangement of the tissue glands in the gastric sinus department,with inflammatory cell infiltration.Conclusion DGP rats model can be established by a single intraperitoneal injection of 2%50 mg/kg STZ combined with high-sugar and high-fat diet for 6 weeks.
