Effect of Wumei pill on intestinal mucosa of ulcerative colitis rats based on PI3K/AKT signal pathway
Objective To observe the repair effect of Wumei pill on intestinal mucosa in rats with ulcerative colitis(UC)based on PI3K/AKT signaling pathway.Methods Rat UC model was established by DSS,and the normal con-trol group were normal rats.The model group,Wumei pill group and Wumei pill+740 Y-P group were all established.After the model was established,Wumei pill group was gavaged with 13.5 g/kg,while Wumei pill+740 Y-P group was intraperitoneally injected with 50 μL of 740 Y-P on the basis of gavage.The colon length,serum levels of D-LA,LPS and DAO were detected,and the expressions of MUC2 and Occludin in colon tissue were detected by immunohistochem-istry.Western blotting was used to detect the expression of PI3K/AKT signaling pathway related proteins in colon tis-sue.Results At the same time,the body mass of rats in the model group was significantly lower than that in the normal control group,and the body mass of rats in the Wumei pill group was significantly higher than that in the model group and Wumei pill+740 Y-P group(all P<0.05).The colon length of rats in the model group was significantly shorter than that in the normal control group,and the percentage of MUC2 and Occludin positive cells was significantly lower than that in the normal control group.The levels of D-LA,DAO and LPS in serum,the score of colon tissue,the rela-tive expression of p-PI3K and p-AKT were higher than those in normal control group.The colon length of rats in Wumei pill group was significantly longer than that in model group and Wumei pill+740 Y-P group,and the percentage of posi-tive cells of MUC2 and Occludin,the relative expression of p-PI3K and p-AKT were significantly higher than that in model group and Wumei pill+740 Y-P group.The levels of serum D-LA,DAO and LPS,and the score of colon tissue were lower than those of model group and Wumei pill+740 Y-P group(all P<0.05).Conclusion Wumei pill can al-leviate the pathological injury of colon tissue and improve the barrier function of colon mucosa,which may be related to the inhibition of PI3K/AKT signaling pathway.
