Proteomic analysis of colorectal cancer and its application
Objective To analyze different proteins in colorectal cancer(CRC)by LC-MS/MS and further screen underling mechanisms related to CRC.Methods LC-MS/MS was used to quantitatively analyze the proteomes of tumor tissues and adjacent normal tissues from 3 CRC patients.Candidate proteins closely related to the occurrence and devel-opment of CRC were screened and verified by bioinformatics analysis.Screening of potential biomarkers of CRC by TCGA analysis.Results A total of 4 257 proteins were identified,including 544 differential proteins,of which 446 were significantly up-regulated and 98 were significantly down-regulated.Based on GO enrichment analysis and KEGG pathway analysis,it was found that proteins related to RNA and DNA functions were involved in the occurrence and de-velopment of CRC by regulating gene expression and cell proliferation.Based on this,the differential proteins related to RNA function were screened as candidate proteins,including PRPF19,HNRNPM,UTP18,RCL1,RPL3,EIF5,NCBP2,NOP58,RANBP2,and their expression was verified in five other CRC patients.It was successfully verified that the expression of proteins NCBP2,PRPF19 and HNRNPM was significantly increased in CRC,and the expression level of NCBP2 was negatively correlated with the overall survival rate of patients.Conclusion This study screened 3 differential proteins with potentially important roles in the occurrence and development of CRC,which are expected to be targets for the diagnosis and treatment of CRC.
