摘要

目的 基于网络药理学和体外细胞实验探究花旗松素治疗卵巢癌的作用机制.方法 查询文献及数据库检索收集花旗松素的作用靶点,采用SuperPred数据库预测靶点,下载疾病基因,找到药物与疾病靶点的交集,进行蛋白质-蛋白质相互作用分析,获取核心靶点,进行富集分析及分子对接;体外培养人卵巢癌SKOV3 细胞,设立空白对照组、花旗松素处理组(10、30 µg/ml),比较各组细胞抑制率及磷酸化丝氨酸/苏氨酸特异性蛋白激酶(p-AKT)、丝氨酸/苏氨酸特异性蛋白激酶(AKT)表达.结果 花旗松素作用于卵巢癌有 79 个潜在靶点和12 个核心靶点.富集分析结果提示,靶点蛋白在癌细胞内异常表达,通过调节代谢等生物过程,影响酶类及蛋白质结合等功能.KEGG主要富集在PI3K/AKT通路,分子对接提示花旗松素与各靶点具有良好的结合活性.体外实验证实,花旗松素可抑制卵巢癌细胞的活性;Western blot结果显示,花旗松素可降低p-AKT/AKT蛋白的表达,从而发挥抗肿瘤作用.结论 花旗松素可通过PI3K/AKT通路抑制卵巢癌细胞的增殖.

Abstract

Objective To explore the mechanism of action of taxifolin in the treatment of ovarian cancer based on network pharmacology and in vitro cell experiments.Methods Literature and databases were searched to collect the targets of action of taxifolin,predict the targets in SuperPred database,download disease genes and find the intersection between drugs and disease targets,perform protein-protein interaction analysis,obtain the core targets,and perform enrichment analysis and molecular docking.Human ovarian cancer SKOV3 cells were cultured in vitro,and the blank control group and the taxifolin treated group(10,30 µg/ml)were set up.The cell inhibition rates and the expression of phosphorylated serine/threonine kinase proteins(p-AKT)and serine/threonine kinase proteins(AKT)of each group were compared.Results There were 79 potential targets and 12 core targets for the action of taxifolin on ovarian cancer.The enrichment analysis results indicated abnormal expression of target proteins in cancer cells,which affected enzyme and protein binding functions by regulating metabolic and other biological processes.KEGG was mainly enriched in the PI3K/AKT pathway,and molecular docking suggested that taxifolin had good binding activity with various targets.In vitro experiments confirmed that taxifolin could inhibit the activity of ovarian cancer cells.Western blot results showed that taxifolin could reduce the expression of p-AKT/AKT protein,thereby exerting anti-tumor effects.Conclusion Taxifolin can inhibit the proliferation of ovarian cancer cells through the PI3K/AKT pathway.
