Effect and Mechanism of Ethyl 3,4-Dihydroxybenzoate from Artemisia capillaris Thunb.Aerial Part on Non-Alcoholic Steatohepatitis
Artemisia capillaris Thunb.aerial part is a kind of food homologous traditional Chinese medicine which can clear away dampness and heat, promote gallbladder and reduce jaundice. In this study, ethyl 3,4-dihydroxybenzoate(EDHB),a component with significant antioxidant activity, was screened from A.capillaris aerial part by H2O2 oxidative stress cell injury model, and then the formation of lipid droplets in fatty liver was simulated by palmitic acid/oleic acid(PA/OA)further verified the anti-non-alcoholic steatohepatitis(anti-NASH)effect of EDHB in vitro.Animal experiment, established by NASH rat model induced by Methionine choline deficiency(MCD)diet, was used to evaluate the effect of EDHB in vivo. Results showed that EDHB from Artemisia capillaris aerial part could effectively alleviate NASH by improving the pathological changes, reducing the steatosis, lowering the transaminase levels,as well as inhibiting the expression and transcription of inflammatory factors including nuclear factor erythroid 2-related factor 2(Nrf2)in liver tissue.
