Sequencing and analysis of the complete mitochondrial genome of Phocodermabetis(Lepidoptera:limacodidae)
Objective:To determine the complete sequences of the mitogenome of Phocodermabetis,and analyze the characteristics of the base composition and codon usage bias of mitogenome,so as to investigate the phylogenetic position of Phocodermabetisin Lepidoptera.Methods:In this study,the complete mitochondrial sequence of Phocodermabetis was sequenced by Illumina high-throughput sequencing technique.Then the mitochondrial sequence were annotated and analyzed using bioinformatics software,including Mitos2,Geneious v8.0.4,and Phylosuite 1.2.2.Finally,two phylogenetic trees were constructed by both maximum likelihoodandBayesianmethods based on 13 protein coding genes of 26 species of Lepidoptera.Results:The complete mitochondrial sequence of Phocodermabetis was 14 919bp(AT:80.88%).The mitogenome consisted of 13 protein coding genes,22 tRNA genes,2 rRNA genes,and an AT enrichment region.The AT content,AT-skew,and GC-skew of the mitogenome of Phocodermabetis showed higher similarity.The phylogenetic trees revealed[Tortricidae+(Sesiidae+Cossidae)+(Limacodidae+Zygaenidae)].Conclusion:The family Limacodidae and the family Zygaenidae represent sister group,belonging to the superfamily Zygaenoidea,and Phocodermabetis is in close genetic relationship with the genus Parasa.
