Healing effect of BMP-2-loaded chitosan hydrogel on the replantation of completely dislocated teeth in rats
Objective:To investigate the osteogenic effect of BMP-2-loaded chitosan hydrogel in the healing process of replanted dislocated teeth in rats.Methods:Twenty-four SD rats were randomly divided into 4 groups.Group A served as blank control group,in which rats were injected with normal saline into the alveolar sockets.Group B and C were experimental controls.Group B received an injection of chitosan hydrogel, and Group C received BMP-2 solution.Group D,the experimental group, was administered with BMP-2-loaded chitosan hydrogel.Under general anesthesia, the right maxillary central incisors of rats were completely extracted, followed by removal of the periodontal ligament and dental pulp.Vitapex was injected into the apical foramen, which was then sealed with glass ionomer. The four groups were injected with the set reagents respectively, and then the teeth were implanted into the alveolar sockets and fixed. After two months, the alveolar bone with maxillary anterior teeth was retrieved, decalcified, embedded, sectioned, and stained with HE.The periodontal ligament healing, osseous healing, and inflammatory resorption in each group of samples were observed under an electron microscope. The data were statistically processed using SPSS 24.0 software. Results:The periodontal ligament healing score of Group D was significantly higher than that of Groups A,B,and C.Its inflammatory resorption score was significantly lower. These differences were also statistically significant(P<0.05).No significant differences were observed in periodontal ligament healing and inflammatory resorption between other groups(P>0.05).Conclusion:BMP-2-loaded chitosan hydrogel can effectively promote the repair of periodontal defects after replantation of dislocated teeth in rats.
