Electroacupuncture treatment regulates the effect of miR-183 on VEGF expression in cochlear hair cells of noise-induced hearing loss model
Objective:To investigate the effect and mechanism of electroacupuncture treatment in regulation of miR-183 on VEGF expression in cochlear hair cells in animal models of noise-induced hearing loss(NIHL).Methods:Male SD rats were randomly divided into control group,NIHL group,NIHL+acupuncture group,NIHL+acupuncture+AAV-NC group and NIHL+acupuncture+AAV-sp-miR-183 group.Rats in the acupuncture were stimulated via Zhongzhu point.MiR-183 was knocked down in NIHL rats using adeno-associated virus(AAV)vectors.The auditory brainstem response(ABR)was evaluated and the hair cells were counted.qRT-PCR was used to analyze the level of miR-183 in rat cochlear tissue.VEGF levels were determined by Western blot.Results:Compared with the control group,the level of miR-183 in cochlear tissue of NIHL group was significantly decreased,whereas the shift of ABR threshold and the loss rate of hair cells as well as the expression level of VEGF in cochlear hair cells were notably increased(all P<0.05).Markedly increased level of miR-183,yet decreased shift of ABR threshold and the loss rate of hair cells and escalated VEGF expression level were seen in cochlear tissue in NIHL+acupuncture group compared to NIHL group(all P<0.05).Comparison of NIHL+acupuncture+AAV-NC group with NIHL+acupuncture+AAV-sp-miR-183 group indicated that miR-183 level was significantly down-regulated,the shift of ABR threshold and the loss rate of hair cells were notably increased,yet VEGF expression level in cochlear hair cells was markedly attenuated(all<0.05).Conclusion:Electroacupuncture could promote VEGF expression in cochlear hair cells of NIHL rats by up-regulating miR-183,and reduce the increase of ABR threshold shift and hair cell damage in the NIHL rat models.
