Objective To explore the effectiveness of domestic disc centrifuges in the separation and purification process of yeast fermentation in large-scale production,and to find out the applicability of domestic disc centrifuge for the separation and purification of yeast fermentation broth.Methods The A600 nm value of supernatant during the centrifugation process was detected to evaluate the effectiveness of separation.The solid content of bacteria after centrifugation was measured to determine whether the centrifugation effect met the requirements for bacterial separation.The possible loss of bacteria in the centrifugation process was measured by calculating the final recovery rate.In order to validate the applicability in production scale,the process duration and bacterial solid content were compared between domestic dish centrifuges and high-capacity centrifuges.As well,morphology of yeast bacterial was compared before and after centrifugation by domestic disc centrifuge,to evaluate the effect of shear force on the separation and purification of yeast.Results There was no significant difference in A600nm value of the supernatant of yeast fermentation between domestic dish centrifuges and high-capacity centrifuges(P=0.053 3,P>0.05).The bacterial solid contents of all batches af-ter centrifugation by domestic dish centrifuges were above 80%,which were close to those after centrifugation by high-capacity centrifuges.No significant difference was observed in the recov-ery of yeast bacteria after centrifugation between domestic dish centrifuge and high-capacity centrifuge(P=0.066 4,P>0.05).In large-scale production,domestic dish centrifuge could shorten the centrifuge process duration significantly(P<0.000 1)compared to large capacity centrifuges.Morphology of yeast bacterial before and after centrifugation in domestic dish centrifuge showed that shear force had basically no impact on yeast morphology.Conclusion In the separation of yeast bacterial fermentation,the centrifugation effect of domestic disc centrifuge is similar to that of the high-capacity centrifuge.However,the process duration is shorter,which is more suitable for large-scale production in this study,and can be expand-ed to large-scale application of other products.
