Objective:To observe and compare effects of chloral hydrate injection anesthesia and isoflurane in-halation anesthesia on the severity of lung tissue and microcirculation in rats with severe acute pancreatitis-associated acute lung injury(SAP-ALI),and to guide the establishment of the SAP-ALI animal model.Method:Thirty male Wistar rats were randomly divided into three groups.Rat models of SAP-ALI were established using chloral hydrate injection(injection group,n=12)and isoflurane inhalation anesthesia inhalation group(inhalation group,n=12),respectively.The remaining 6 rats were used as controls(control group).The 24-hour mortality rate and gross pathological changes in intraperitoneal tissues were observed.The concentration of serum amylase was detected by enzyme-linked immunosorbent assay(ELISA).The changes in pulmonary blood flow in rats were detected using Moor blood flow and oxygen monitor and laser confocal blood flow and oxygen monitor.Pathological changes and differences in lung and pancreatic tissues were observed.Results:Both anesthesia methods successfully established rat models of SAP-ALI.However,compared with the inhalation group,the rats in the injection group had higher mortality rates,lower pulmonary microvascular blood flow(P<0.05),and more severe pathological manifestations in the lung tissue(P<0.05).Conclusion:It is recommended to use isoflurane inhalation anesthesia when establis-hing rat models of SAP-ALI to improve the accuracy of experimental results.
