Optimization of Decolorization Process and Antioxidant Activity of Crude Polysaccharides from Bo Baizhi Using Response Surface Macroporous Resin Method
In order to study the decolorization process and antioxidant activity of Bozhong crude polysaccharide macroporous resin.Using the comprehensive evaluation of decolorization rate and polysaccharide retention rate as indicators,an experiment was designed using response surface methodology to optimize the decolorization process of crude polysaccharide macroporous resin from Radix Angelicae Dahuricae;And the scavenging ability of DPPH and hydroxyl radicals was measured to study the antioxidant activity of crude polysaccharides from Angelica dahurica before and after decolorization.The results showed that the optimal parameters for the decolorization process of crude polysaccharides from Angelica dahurica were:the mass ratio of crude polysaccharides to D-301 macroporous resin wet weight was 1:31,decolorization temperature was 60 ℃,decolorization time was 3.2h,decolorization pH value was 7.3,and the final decolorization rate of polysaccharides was 89.03%,retention rate was 82.14%,and comprehensive score was 85.59%;After decolorization,the DPPH and hydroxyl radical scavenging rates of polysaccharides were slightly lower than before decolorization,and the concentrations were close to the VC positive control group when they were higher than 4.0mg/mL and 3.0mg/mL,respectively.Conclusion:The D301 macroporous resin decolorization process has strong stability and feasibility,and can be applied to practical production research;After decolorization,polysaccharides from Baizhi have good antioxidant activity and can be used as natural antioxidants.
