昆明葡萄酒相关酵母菌的分离与鉴定

[目的]分离鉴定云南昆明葡萄产区的葡萄酒相关酵母,为该地葡萄酿酒酵母资源的筛选及其利用奠定基础.[方法]以采自昆明官渡区三瓦村的葡萄桨果为材料,将葡萄果皮用灭菌的液体SelMed富集培养基进行培养,经稀释再培养以获得酵母单菌落,采用ITS序列分析和RFLP鉴定分离的相关菌种,并利用微卫星指纹图谱对酿酒酵母的不同菌株进行鉴定.[结果]从云南昆明葡萄产区葡萄果皮上分离获得了207个单克隆菌落.PCR扩增和RFLP分析表明,这些单菌落分属于3个属,分别为梅奇酵母属(Metschnikowia)、红酵母属(Rhodotorula)和酿酒酵母属(Saccharomyces),其中梅奇酵母属出现频率最高,并有23个酿酒酵母单菌落.微卫星分析结果表明,23个酿酒酵母菌落分属于4个菌株,其中1个为商业菌株X5.[结论]在昆明的葡萄园中至少栖息了3个属5个种的酵母,包括酿酒酵母(Saccharom yces cerevisiae)、胶红酵母(Rhodotorula mucilaginosa)、金佩梅奇酵母(Metschnikowi chrysoperlae)、美极梅奇酵母(Metschnikowi pulcherrima)和核果梅奇酵母(Metschnikowi fructicola)等.分离到的葡萄酒相关酵母部分源于欧洲,可能随着葡萄植株被引进到昆明.
Isolation and identification of wine related yeasts in Kunming
[Objective] This study focused on the isolation and identification of grape wine related yeasts in Kunming,Yunnan Province,to improve the screening of wine related yeasts and their subsequent use.[Method] Using grape fruits from Sanwa village,Guandu District,Kunming,sterile liquid SelMed enrichment medium was used to cultivate the yeasts from grape skin and monoclonal yeasts were obtained after dilution and culturing.ITS sequence analysis and RFLP analysis were used to identify related species,and microsatellites were used to access the different strains of Saccharomyces cerevisiae.[Result] 207 monoclonal isolates fingerprints were obtained from grape skin of Kunming,Yunnan.PCR and RFLP analysis revealed that these colonies belonged to 3 genera,Metschnikowia,Rhodotorula,and Saccharomyces.Metschnikowia was the richest genera,and 23 colonies belonged to S.cerevisiae.Microsatellite analysis showed that these S.cerevisiae colonies belonged to four strains,and one was commercial strain X5.[Conclusion] In vineyards of Kunming,there were at least five yeasts belonging to 3 genera,including S.cerevisiae,Rhodotorula mucilaginosa,Metschnikowia chrysoperlae,Metschnikowia pulcherrima,and Metschnikowia fructicola.Some of the isolated yeasts originated from Europe,which may be introduced to Kunming together with the grape plants.
