Preparation of SLF-1 microbial agent and its effect on rape growth
The rational use of microbial agents can improve the quality of agricultural products and reduce the harm to soil and environment caused by excessive use of chemical fertilizers and pesticides in agriculture.In this study,one strain of endophytic Bacillus mycoides SLF-1 isolated and screened from the root nodules of Hippophae rhamnoides was used for microbial agent.The strain SLF-1 has antibacterial and growth promoting abilities.Its effect on the growth of Brassia campestris was tested through potted and field experiments.The quality of SLF-1 bacterial agent was detected using dilution coating method.The single factor experiments were used to optimize the types and concentrations of carbon and nitrogen sources using beef extract peptone liquid medium as the base culture medium.The results showed that the strain SLF-1 grows best when 5 g·L-1 malt starch is used as the carbon source and 10 g·L-1 soybean meal as the nitrogen source for liquid fermentation.the strain SLF-1.It not only meets the cost-effectiveness of raw materials for large scale production of microbial agents,but also achieves the best growth effect of bacterial cells.The SLF-1 bacterial content in the microbial agent can reach 69.2%of the fermentation culture using beef extract peptone medium.The number of live bacteria meets the national standard for agricultural microbial agents.The application of SLF-1 microbial agent has a promoting effect on the fresh weight,plant height,leaf length,leaf width,and chlorophyll content of B.campestris in pot and field experiments.In field experiments,B.campestris grows stronger due to the application of SLF-1 microbial agents.
