桂中报春苣苔花粉生物学特征与繁育系统研究

[目的]探究桂中报春苣苔花粉的生物学特征与繁育系统对该物种有性生殖的影响,揭示其繁殖机制,为报春苣苔属植物的保护和开发工作提供理论参考.[方法]以桂中报春苣苔为材料,观察其花部特征,筛选花粉离体萌发培养基及最佳的花粉活力检测方法,探索最佳的花粉贮藏条件,判断其繁育系统类型.[结果](1)桂中报春苣苔雌雄异熟,雄蕊先熟于雌蕊;雌雄异位,雌蕊高于雄蕊;(2)花粉离体萌发培养的最优培养基配方为40 mg/mL硼酸+10%蔗糖+7 g/L琼脂,培养时间为2 h;(3)花粉在-20 ℃和-80 ℃条件下贮藏7 d后均能保持较高的萌发率;(4)柱头在开花2~4 d可授性最强;(5)花粉胚珠比(P/O)为518.36~782.75,杂交指数(OCI)为5;(6)有较高的自交亲和性,不能进行无融合生殖.[结论]离体萌发法是最为直观准确的花粉活力检测方式,低温有利于桂中报春苣苔花粉贮藏,繁育系统以异交为主.
Pollen biological characteristics and breeding system of Primulina guizhongensis
[Objective]This paper aims to investigate the characteristics of Primulina guizhongensis pollen and the influence of the breeding system on its sexual reproduction,to uncover its reproduction mechanism and providing theoretical references for the conservation and development of Primulina.[Methods]Using P.guizhongensis as the material,we observed the floral characteristics of P.guizhongensis,screened the pollen isolation medium and the best pollen viability detection method,explored optimal pollen storage conditions,as well as determined the type of its breeding system.[Results](1)P.guizhongensis was di-chogamy,with stamens ripening before pistils.It was herkogamy,with pistils higher than stamens.(2)The optimum medium for culture of the isolated pollen germination was 40 mg/mL boric acid+10%su-crose+7 g/L agar,for two hours.(3)Pollen maintained a high germination rate after 7 d of storage at both-20 ℃ and-80 ℃.(4)The stigmas of P.guizhongensis were most fertile at 2-4 d of flowering.(5)P/O was 518.36-782.75,and OCI was 5.(6)P.guizhongensis had a high self-fertilization affinity and was unable to undergo fusionless reproduction.[Conclusion]In vitro germination method is the most intuitive and accurate way to detect pollen viability.Low temperature is conducive to pollen storage of P.guizhongensis,and the breeding system is predominantly heterogametic.
