摘要
以卵磷脂和胆固醇为壁材,采用乙醇注入法制备EGCG(表没食子儿茶素没食子酸酯)脂质体.系统考察了制备过程中壁材质量比、乙醇加入量、EGCG添加量、搅拌温度、吐温80用量、搅拌时间对脂质体粒径和包封率的影响,并通过正交实验分析得出最优组合.采用纳米粒径分析仪、紫外分光光度计等对制备的脂质体微胶囊进行表征.结果表明,当胆固醇:卵磷脂质量比为1∶5、EGCG质量分数为0.06%、吐温80质量分数为0、乙醇体积分数为14%、搅拌温度为40℃、搅拌速度为540 r/min时,脂质体粒径小于100 nm,包封率大于90%.通过实验优化,能够得到小粒径、高包封率的脂质体,提高了 EGCG生物利用度.
Abstract
EGCG(epigallocatechin gallate)liposomes are prepared via ethanol injection with lecithin and cholesterol as wall materials.The influences of the mass ratio of wall materials,the dosage of ethanol,the dosage of EGCG,the stirring temperature,the usage of Twain 80 and the stirring time on the particle size and the encapsulation rate of liposomes are evaluated systematically,and the optimal combination is obtained through orthogonal experimental analysis.The prepared liposome microcapsules are characterized by means of nanometer particle size analyzer and ultraviolet spectrophotometer.Results show that the particle size of liposomes is less than 100 nm and the encapsulation rate exceeds 90%when the mass ratio of cholesterol to lecithin is 1∶5,the mass fraction of EGCG is 0.06%,the mass fraction of Tween 80 is 0,the volume fraction of ethanol is 14%,the stirring temperature is 40℃ and the stirring speed is 540 r·min-1.Through experimental optimization,the liposomes with small particle size and high encapsulation rate can be obtained,and the bioavailability of EGCG can be improved.
