目的 应用生物信息分析和实验验证探讨转录因子性别决定区Y-box 6(SOX6)与转录因子性别决定区Y-box 17(SOX17)在口腔鳞状细胞癌(OSCC)中的表达及意义.方法 获取癌症基因组图谱(The Cancer Genome Atlas,TCGA)下载的OSCC组织及相应的癌旁非瘤组织的转录组数据,分析SOX6、SOX17的表达水平与OSCC发生发展及其预后的相关性.收集口腔鳞状细胞癌组织及相应的癌旁非瘤组织标本,应用免疫组化SP法分析SOX6与SOX17在OSCC中的表达及其相关性.利用蛋白-蛋白互作(PPI)网络、基因本体论(GO)分析、京都基因与基因组百科全书(KEGG)通路富集分析探讨与SOX6相关的基因、富集通路及其对OSCC发生发展的影响.结果 生物信息分析显示,在OSCC组织中SOX6、SOX17的表达水平显著低于癌旁组织(P<0.05),SOX6、SOX17在OSCC中有较高的诊断价值(曲线下面积(AUC)分别为0.972、0.731),且其高表达与患者的良好预后相关,Cox回归分析显示,TNM分期、SOX6、SOX17表达水平是OS的独立预后因素(P<0.05).免疫组化分析显示,SOX6在OSCC组织中含量与肿瘤浸润深度密切相关(P<0.05),Spearman分析显示,SOX6、SOX17表达水平呈正相关(r=0.297,P<0.05).通过PPI网络获得互作蛋白,GO与KEGG结果显示SOX6可能与FAK信号通路、Notch信号通路和肿瘤细胞的增殖迁移等关系密切.结论 SOX6、SOX17在OSCC组织中呈低表达,是OSCC潜在的诊断及预后标记物.
Expression and clinical significance of SOX6 and SOX17 in oral squamous cell carcinoma
Objective Bioinformatics analysis and experimental validation were used to investigate the expression and significance of transcription factor Sextinite region Y-box 6(SOX6)and transcription factor Sextinct region Y-box 17(SOX17)in oral squamous cell carcinoma(OSCC).Methods The transcriptome data of OSCC tissues and corresponding adjacent non-tumor tissues downloaded from The Cancer Genome Atlas(TCGA)were obtained,and the correlation between the expression levels of SOX6 and SOX17 and the occurrence and development of OSCC and its prognosis was analyzed.Oral squamous cell carcinoma tissues and corresponding adjacent non-tumor tissue specimens were collected,and the correlation between SOX6 and SOX17 expression was analyzed by immunohistochemistry SP method.Protein-protein interaction(PPI)network,gene ontology(GO)analysis,Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were used to explore the genes and enrichment pathways related to SOX6 and their effects on the occurrence and development of OSCC.Results Bioinformatics analysis showed that the expression levels of SOX6 and SOX17 in OSCC tissues were significantly lower than those in adjacent tissues(P<0.05),and SOX6 and SOX17 had higher diagnostic value in OSCC(area under the curve(AUC)were 0.972 and 0.731,respectively),and their high expression was associated with a good prognosis of patients,and Cox regression analysis showed that TNM stage,SOX6 and SOX17 expression levels were independent prognostic factors for OS(P<0.05).Immunohistochemical analysis showed that the content of SOX6 in OSCC tissues was closely related to the depth of tumor invasion(P<0.05),and Spearman analysis showed that the expression levels of SOX6 and SOX17 were positively correlated(r=0.297,P<0.05).The results of GO and KEGG showed that SOX6 may be closely related to FAK signaling pathway,Notch signaling pathway,and the proliferation and migration of tumor cells.Conclusion SOX6 and SOX17 were lowly expressed in OSCC tissues,which were potential diagnostic and prognostic markers of OSCC.
