Expression and significance of NEFL in oral squamous cell carcinoma
Objective To investigate the expression of NEFL(neurofilament light chain)in oral squamous cell carcinoma(OSCC),to study the effect of NEFL on cell proliferation,invasion and migration of oral squamous cell carcinoma,and to explore the potential molecular mechanisms.Methods The expression of NEFL in pan-cancer as well as OSCC was analysed based on OSCC clinical data from The Cancer Genome Atlas(TCGA)data.Also,the effect of NEFL on different clinical factors in OSCC was analysed.Immunoprotein blotting(Western Blot)assay was performed to detect the expression of NEFL in oral squamous carcinoma tissues and paracancerous tissues as well as the expression of NEFL in OSCC cell lines and normal oral mucosal keratinocytes(HOK).The effects of NEFL on the proliferation,migration and invasion ability of OSCC cells were detected by CCK8 assay,EDU assay,scratch assay and Transwell assay after knocking down NEFL,and Western Blot was used to detect the effects of NEFL on the PI3K-AKT signaling pathway.Results The results showed that NEFL was highly expressed in OSCC and other tumors,and had good diagnostic significance(AUC:0.820).Knockdown of NEFL could inhibit the proliferation,invasion and migration of OSCC cells.The expression of p-PI3K and p-AKT in OSCC cells was also reduced after NEFL knockdown.Conclusion Low expression of NEFL regulates the PI3K-AKT signaling pathway by inhibiting the phosphorylation of PI3K and AKT,thereby inhibiting the proliferation,invasion and migration of OSCC cells,and thus participating in the malignant progression of OSCC.
