Discussion on the test method of sample carry-over in liquid chromatograph autosampler
The sample carry-over of liquid chromatograph autosampler was very important to the test results of liquid chromatography.According to the test conditions of GB/T 38125-2019,naphthalene was used as the sample,and both the mobile phase and the sample solvent were methanol,which would produce an interference peak near the main peak position,affecting the accuracy of the sample carry-over test results.Therefore,in this paper,by using caffeine because of its good solubility in both aqueous and organic phases,different sample solvents and different proportions of mobile phase were used for sample carry-over test,and the sample carry-over testchromatogram under different chromatographic conditions were compared.Through analysis and discussion,when the mobile phase was 70%water/30%methanol,and the sample solvent was 100%water or 70%water/30%methanol,the interference peak area could be reduced and the interference peak and the main peak could be separated obviously,so as to have no impact on the sample carry-over test.Among them,70%water/30%methanol as mobile phase and 100%water as sample solvent were the better conditions for sample carry-over test in autosampler.
