Comparison of four different biomolecular interaction detection techniques
The interactions between biological molecules,such as protein and protein,protein and ligand,nucleic acid and ligand form the basisof life activities.The study of the interaction can clarify the mechanism of biological reactions and reveal the nature of life activities from the molecular level.In development of innovative drugs,the study of the interaction between drug molecules and target proteins runs through the entire process of drug development,which is of great significance for understanding the mechanism of action of drugs and guiding the optimization and improvement of drugs.This article will introduce four of the most commonly used biophysical techniques for studying interactionsbased on different technical principles,including isothermal titration calorimeter(ITC),surface plasmon resonance(SPR),biolayer interferometry(BLI),and microscale thermophoresis(MST).Combined with practical cases to comprehensively compare the characteristics of the four techniques,theapplication of each techniques in the field of biomedicine will be discussed in detail,aiming to provide references for the selection of interaction detection methods in the research process.
