微滴数字PCR定量检测蔬菜中大肠埃希氏菌O157:H7/NM

建立了蔬菜中大肠埃希氏菌O157:H7/NM(Escherichia coli O157:H7/NM)微滴数字聚合酶链式反应(微滴数字PCR)快速精准定量检测方法,评价其可行性。采用该菌rfbE 基因的特异性引物和探针,建立微滴数字PCR体系。微滴数字PCR法检测大肠埃希氏菌O157:H7/NM的检测限低至 1×103CFU/mL纯菌液,且该方法具有较高的特异性和敏感性,重复性试验结果稳定,不与其他常见病原菌发生交叉反应。本研究建立了微滴数字PCR法检测蔬菜中大肠埃希氏菌O157:H7/NM的定量方法。
High-sensitivity quantitative determination of Escherichia coli O157:H7/NM in vegetable by Droplet Digital PCR
A rapid and accurate method for the detection of Escherichia coli O157:H7/NM by microdrop digital polymerase chain reaction(droplet digital PCR)in vegetables was established to evaluate its feasibility.A microdrop digital PCR system was established by using specific primers and probes of rfbE gene.The detection limit of microdrop digital PCR for Escherichia coli O157:H7/NM was as low as 1×103 CFU/mL pure bacterial solution,and this method had high specificity and sensitivity,stable repeatability results,and did not cross-react with other common pathogens.A quantitative method for detection of Escherichia coli O157:H7/NM in vegetables by microdrop digital PCR was established.
