肌层浸润性膀胱癌中CD276的表达与临床病理特征和预后的相关性

摘要
目的 CD276 的异常表达已被证实与肿瘤的发生有关.但其在肌层浸润性膀胱癌(MIBC)中的作用知之甚少.方法 从癌症基因组图谱数据库中下载 MIBC患者的相关数据,通过研究CD276 在膀胱正常组织和肿瘤组织中的表达差异,同时评估临床变量与 CD276 之间的相关性来预测CD276 与MIBC的生存预后.采用Cox回归分析和人类蛋白质图谱证明CD276 是MIBC的独立预测因子.最后使用基因集富集分析(GSEA)数据集进行分析.结果 MIBC 的肿瘤分级、分期、发病年龄和 N分期与CD276 的高表达有关.CD276 高表达患者预后较 CD276 低表达者差.单因素和多因素回归分析显示,CD276 是MIBC的独立危险因素.GSEA富集分析发现CD276主要富集于肿瘤相关通路、膀胱癌、细胞黏附分子、ECM受体相互作用、Wnt信号通路、趋化因子信号通路以及 MAPK信号通路.结论 CD276 在 MIBC中高表达,其可作为 MIBC预后的潜在独立分子标志物.
Abstract
Objective The abnormal expression of CD276 has been shown to be associated with tumorigenesis.However,its role in muscle-invasive bladder cancer(MIBC)is poorly under-stood.Methods The relevant data of MIBC patients were downloaded from the TCGA database,then correlation between CD276 and the survival prognosis of MIBC was predicted by studying the differential expression of CD276 in normal and tumor tissues of the bladder and assessing the correla-tion between clinical variables and CD276.Cox regression analysis was used to demonstrate that CD276 was an independent predictor of MIBC.Further validation was performed using the Human Protein Atlas.Gene set enrichment analysis GSEA was finally performed.Results The tumor grade,stage,age of onset and N stage of MIBC were related to the high expression of CD276.Pa-tients with high CD276 expression had a worse prognosis than those with low CD276 expression.Univariate and multivariate regression analysis showed that CD276 was an independent risk factor in MIBC.GSEA revealed that CD276 was mainly enriched in pathways in cancer,bladder cancer,cell adhesion molecules,ECM receptor interactions,Wnt signaling pathway,chemokine signaling path-way as well as MAPK signaling pathway.Conclusions We confirmed the high level expression of CD276 in MIBC,which can be used as a potential independent molecular marker for the prognosis of MIBC.
