Objective To study the effects of Berberine on the growth of human bladder cancer cell lines 5637,and to elucidate the possible mechanism of such effects.Methods The inhibitory effect of Berberine on cell proliferation was examined by CCK-8 and cell cycle was detected by flow cytometry(FCM).The expression levels of CDK1,EZH2 mRNA and protein levels were detected by qPCR and Western blotting.Results The Berberine significantly inhibited cell proliferation,and blocked the cell cycle in the G0/G1 phase.The expression levels of CDK1,EZH2 mRNA and protein down-regulated when 5637 cell were treated by Berberine.Conclusions Berberine signifi-cantly inhibited cell proliferation,and blocked the cell cycle in the G0/G1 phase.These effects may be related with its function in down-regulating the expression levels of CDK1 and EZH2 protein.
