Objective To investigate the effect of Huaier aqueous extract on the proliferation and migration of bladder cancer cells.Methods CCK-8 method and plate cloning were applied to detect the proliferation ability of bladder cancer cells,and Transwell assay was used to detect the in-vasive and migratory ability of bladder cancer cells.Western blot was used to detect the expression levels of matrix metalloproteinase-2(MMP-2),matrix metalloproteinase-9(MMP-9),Vimentin,N-cadherin and E-cadherin in bladder cancer cells.Results The results of CCK-8 showed that the cell viability of RT4 and MB49 cells was significantly inhibited in a concentration and time dependent manner after treating with low concentration(3 mg/ml),the middle concentration(6 mg/ml),and high concentration(12 mg/ml),of Huaier aqueous extract,respectively,for 24、48、72 h(P＜0.05).The results of proliferation-invasion assay showed that compared with the control group,the proliferation,invasion and migration ability of bladder cancer cells were reduced by the low,middle and high concentrations of Huaier aqueous extract(P＜0.05).The results of Western blot showed that compared with the control group,the expression levels of MMP-2,MMP-9,Vimentin,and N-cadherin reduced,while the expression level of E-cadherin protein increased in the cells of the low,middle and high concentration groups.There was a significant difference between the groups(P＜0.05).Conclusions Huaier aqueous extract can inhibit bladder cancer cells proliferation,migra-tion,and invasion by altering the expression of epithelial mesenchymal transition-related protein lev-els in bladder cancer cells.
