Objective To explore the expression of lncRNA double homeobox A pseudogene 8(DUXAP8)in prostate cancer and its function in the progression of prostate cancer.Methods The DUXAP8 expression in prostate cancer tissues,and the relationship between DUXAP8 expres-sion and clinical parameters were performed using TCGA dataset.QRT-PCR was used to detect the expression of DUXAP8 in prostate cancer cell lines.DUXAP8 knockdown cell lines were constructed using shRNA.Then transwell,wound healing and EdU assays were used for cells migration and pro-liferation analysis.In vivo xenograft assay was also employed to evaluate the DUXAP8 role in pros-tate cancer.Results We found that DUXAP8 overexpressed in prostate cancer,and its overex-pression was associated with poor progression-free survival,T stage,lymph node metastasis,Glea-son score and pathological T stage(P=0.040,P=0.003,P=0.0240,P=0.009,P=0.035).DUXAP8 knockdown inhibited the prostate cancer cell(PC-3 and DU145)migration and invasion,and cell growth as well(P＜0.001).Conclusions As a pro-oncogene,DUXAP8 may be a poten-tial biomarker for poor prognosis and therapeutic target in prostate cancer.
