HPLC法测定不同制法的四逆汤中3种苯甲酰类乌头碱及6-姜酚含量

目的 建立高效液相色谱法同时测定四逆汤中苯甲酰类乌头碱、6--姜酚的含量,比较3种方法制备的四逆汤中有效成分含量差异.方法 苯甲酰类乌头碱的测定采用C18色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.1 mol.L-1醋酸铵溶液为流动相进行梯度洗脱,流速为1.0 mL·min-1,检测波长为235 nm,柱温30℃.6-姜酚测定采用C18色谱柱(4.6 mm×250 mm,5μm)以乙腈-甲醇-水(40:5:55)为流动相等度洗脱,流速为1.0 mL.min-1,检测波长为280 nm,柱温25℃.结果 3种不同方法提取液中苯甲酰类乌头碱的提取率依次为:有旋部位组合法>药典法>传统法.6-姜酚的提取率依次为:传统法>药典法>有效部位组合法.结论 所建立的方法简单、准确,为四逆汤提取方法改进提供科学依据.
Objective To establish an HPLC method for simultaneous determination of benzoyl aconitine and 6-gingerol in Sini decoction and compare the content differences of active ingredients in Sini decoction prepared by three extraction methods.Methods The detection method of the benzoyl aconitine was conducted on an C18 (4.6 mm× 250 mm,5un) column with acetonitrile-0.1 mol.L-1 ammonium acetate solution as the mobile phase using gradient elution.The flow rate was set at 1.0 mL·min-1 and detection wavelength was set at 235 nm.The column temperature was at 30 ℃.The detection method of 6-gingerol was conducted on an C18 (4.6 mm× 250 mm,5 μm) column with acetonitrile-methanol -water (40:5:55) as the mobile phase using isocratic elution.The flow rate was set at 1.0mL·min1 and the detection wavelength was set at 280 nm.The column temperature was at 25 ℃.Results The extraction rate ofbenzoyl aconitine by different methods decreased in the follow order:the combination of active ingrediets > pharmacopoeia method >traditional method.The extraction rate of 6-gingerol decreased in the follow order:traditional method >pharmacopoeia method > the combination of active ingredients.Conclusion The established method is simple,accurate and can be used for providing a scientific basis for the improvement of extraction method of Sini decoction.
