目的 优化大鼠光化学法脑梗死的实验方法,制备稳定的脑梗死继发脊髓束华勒氏变性大鼠模型并进行标准化评估.方法 通过光化学法建立SD大鼠大脑右侧运动皮层终末动脉永久性缺血性损伤模型(实验组).第一阶段实验组根据 45 只SD大鼠质量分为 3 组:A组,150~250g;B组,250~350g;C组,350~450g.每组再根据 5%玫瑰红B剂量随机平均分3个亚组:Ⅰ组,12.5mg/100g体重;Ⅱ组,20mg/100g体重;Ⅲ组,30mg/100g体重.第二阶段实验组在BⅡ组基础上,根据玫瑰红B的浓度分为 3 组:a组,5%;b组,7.5%;c组,10%;每组再根据注射时间随机平均分 3 个亚组:1 组,注射时间<1min;2 组,注射时间 1~2min;3 组,注射时间>2min.造模后 24h扫描MRI,28d扫描扩散峰度成像(Diffusional kurtosis imaging,DKI)后牺牲大鼠,取脑组织做常规HE、髓鞘LFB及免疫荧光GFAP染色.结果 脑梗死继发脊髓束华勒氏变性大鼠模型制备的最佳参数组合为:SD大鼠体重 250~350g、5%玫瑰红B 20mg/100g体重、注射时间≥2min、冷光源探头距离暴露的颅骨大约 20mm、照射强度 300~320klx、照射时间 30min,该参数组合制备模型的 24h脑梗死阳性率、28d脑软化灶阳性率及 28d存活率均为 100%.结论 本研究详细描述了光化学法脑梗死继发脊髓束华勒氏变性大鼠模型建立过程并进行标准化评估,为神经变性疾病研究提供了一种稳定、可靠的动物模型.
Preparation and standardized evaluation of Waller's degeneration rat model of corticospinal tract
Objective The experimental method of photochemical cerebral infarction in rats was optimized,and a stable rat model of Waller's degeneration of spinal cord secondary to cerebral infarction was established and standardized.Methods The model of permanent ischemic injury of terminal artery in right motor cortex of SD rats was established by photochemical method(study group).In the first stage,the study group was divided into three groups according to the mass of 45 SD rats:group A 150~250 g;group B 250~350 g;group C 350~450 g.Each group was randomly divided into 3 subgroups according to 5%rose red B dose:group Ⅰ 12.5mg/100g,group Ⅱ20mg/100g,group Ⅲ30mg/100g.The second stage study group was divided into 3 groups according to the concentration of rose red B on the basis of group B Ⅱ:Group a 5%,Group b 7.5%,Group c 10%.Each group was randomly divided into 3 subgroups according to the average injection time:Group 1 injection time<1 minute,group 2 injection time 1~2 minutes,group 3 injection time>2 minutes.MRI scans were performed 24 hours after modeling,and rats were sacrificed after DKI scans on 28 days.Brain tissues were taken for routine HE,myelin LFB,and immunofluorescence GFAP staining.Results SD rats weighing 250~350g,5%Rose Bengal B(20mg/100g body weight),injection time≥2min;place the cold light source probe about 20mm away from the exposed skull,irradiation intensity 300~320klx,irradiation time 30min,brain The 24-hour cerebral infarction positive rate,the 28-day encephalomalacia positive rate and the 28day survival rate of the rat model of Wallerian degeneration of the spinal cord secondary to infarction were all 100%.Conclusion This study describes in detail the establishment process of the rat model of Wallerian degeneration of the spinal cord secondary to photochemical cerebral infarction and conducts standardized evaluation,providing a stable and reliable animal model for neurodegeneration research.
