Differenceanalysis and prediction of drug resistance value of miR-15a-5p in patients with breast cancer resistant and sensitive groups
Objective To investigate the expression difference of miR-15a-5p in docetaxel sensitive group and drug-resistant group and its significance for clinical prognosis.Methods TCGA database was used to analyze the expression of miR-15a-5p in malignant tumors.30 docetaxel-resistant breast cancer patients treated in the hospital from 2019 to 2020 were collected as the drug-resistant group,and 30 drug-sensitive breast cancer patients treated in the same period were collected as sensitive group,and the expression level of miR-15a-5p in cancer tissues of thepatients were detected by q-PCR fluorescence quantification method.To investigate the association between miR-15a-5p and drug resistance and typing of breast cancer.ROC curve was drawn to analyze the predictive value of miR-15a-5p on patient prognosis and the diagnostic value of miR-15a-5p on drug resistance in breast cancer patients.Results Pan-cancer analysis showed that miR-15a-5p was highly expressed in breast cancer and other cancers.q-PCR results showed that the expression level of miR-15a-5p in docetaxel chemotherapy sensitive group was higher than that in drug resistant group(P<0.05).Clinical statistical analysis showed that miR-15a-5p was negatively correlated with the expres-sion of ER(estrogen receptor)and PR(progesterone receptor),but had no significant correlation with the expression of human epidermal growth factor receptor-2.Conclusion The expression level of miR-15a-5p in tumor tissues of docetaxel resistant patients is significantly lower than that of sensitive group,and the high expression of miR-15a-5p in breast cancer is closely related to the molecular typing and prognosis of breast cancer.MiR-15a-5p may be involved in immune regulation of breast cancer and ultimately lead to docetaxel resistance in breast cancer patients.
