Expression of CircRNA_000121 in papillary thyroid microcarcinoma(PTMC)with cervical lymph node metastasis
Objective To investigate the expression of CircRNA_000121 in papillary thyroid microcarcinoma(PTMC)with cervical lymph node metastasis.Methods 60 patients admitted to the hospital from January to April 2022 were selected as study objects.Among the patients,there were 30 patients with PTMC ac-companied by cervical lymph node metastasis(central region)(PTMC(L)group)and 30 patients with PTMC without lymph node metastasis(PTMC group).Preoperative peripheral blood samples,intraopera-tive thyroid cancer tissues and general data of all patients were collected.The expression levels of circRNA_000121,miR-146b-3p and MMP2 in blood and tissue samples of the patients in the 2 groups were detected by qRT-PCR.Receiver operating characteristic curve(ROC)was used to analyze the diagnostic value of circRNA_000121 in thyroid micropapillary carcinoma with cervical lymph node metastasis.Binary Logistic regression was used to analyze the correlation between the expression levels of circRNA-000121 and miR-146b-3p,miR-146b-3p and MMP2 in peripheral blood samples and tissue samples of PTMC and lymph node metastasis.Results qRT-PCR results showed that compared with PTMC group,circRNA_000121 and MMP2 were highly expressed in peripheral blood and tissue samples of the patients in PTMC(L)group and miRNA-146b-3p was low,with statistical significance(P<0.05).ROC analysis showed that the area under the curve(AUC)of peripheral blood was 0.964,the confidence interval(95%CI)was 0.915～1.000,the sensitivity was 0.933,and the specificity was 1.000.The AUC of tissue specimens was 0.943,the confidence interval(95%CI)was 0.874～1.000,the sensitivity was 0.900,and the specificity was 0.967.Binary Logistic regression analysis showed that the expressions of CircRNA_000121,miRNA-146b-3p and MMP2 in peripheral blood and cancer tissues were correlated with lymph node metastasis,and the differences were statistically significant(P<0.05).Conclusion CircRNA_000121 is highly expressed in thyroid cancer tissue samples of the patients with PTMC(L),which is consistent with the expression in peripheral blood.CircRNA_000121 plays a certain role in predicting lymph node metastasis of PTMC,and it is expected to be a biological marker for cervical lymphatic metastasis of PTMC.
