Objective Study on the preparation method of sodium estrone sulfate(ES)and its protective effect on myocardial ischemia-reperfusion injury in mice.Methods(1)ES was synthesized using estrone and amino sulfonic acid via amino sulfonation and ion displacement methods.The purity of ES was calcu-lated using nuclear magnetic resonance internal standard method,and the structure of ES was analyzed u-sing UV spectroscopy,infrared spectroscopy,high-resolution mass spectrometry,nuclear magnetic reso-nance hydrogen spectroscopy and nuclear magnetic resonance carbon spectroscopy.(2)After establishing a menopausal model in female mice,they were divided into a solvent group(distilled water 0.094 mg/kg body weight),an estradiol group(dose of 0.3 mg/kg),and a sodium estrone sulfate group(dose of 0.094 mg/kg).After 2 weeks of oral administration,a mouse model of myocardial ischemia/reperfusion injury was estab-lished.The infarct area was measured using 2,3,5-triphenyltetrazolium chloride(TTC)staining method to evaluate the effect of ES pretreatment on mouse myocardial injury.Results The synthesized ES has a purity of 66.6%and a Tris content of 30.8%.The characterization results show that the product is sodi-um estrone sulfate.Compared with the solvent group,the ES group mice showed a statistically significant decrease in myocardial infarction area caused by myocardial ischemia/reperfusion injury(P<0.05).Conclu-sion This synthesis method can obtain high purity and stable ES,and animal experiments have shown that ES can reduce the myocardial infarction area after myocardial ischemia-reperfusion injury in mice.
