不同功能化合物对迁移体形成的影响研究

目的 探讨SF1670、秋水仙碱、二甲双胍、阿司匹林等4种不同功能化合物对迁移体形成的影响.方法 Vigofect化学转染法构建L929过表达Tspan4-mCherry细胞系,分为空白对照组(不含任何药物的完全培养基)、SF1670组(4 μmol/L)、秋水仙碱组(0.6 μmol/L)、二甲双胍组(32 µmol/L)和阿司匹林组(1 μmol/L),分别作用11 h;活细胞成像实验拍摄细胞产生的迁移体;Fiji-Image J统计迁移体产生数量,收缩纤维形成面积,直径及长度.结果 共聚焦成像结果显示,L929过表达T span4-mCherry细胞系建立成功,产生了收缩纤维和迁移体.4种药物对迁移体影响结果显示,与空白对照组比较,SF1670组迁移体产生数量减少(P<0.01),收缩纤维的形成面积和长度被明显抑制(P<0.000 1);秋水仙碱组迁移体产生数量明显减少(P<0.000 1),收缩纤维的形成面积轻微减少(P<0.05);二甲双胍组细胞收缩纤维形成面积(P<0.05)和直径(P<0.000 1)均减少;阿司匹林组细胞迁移体产生数量增加(P<0.01),收缩纤维的直径轻微缩短(P<0.05).结论 SF1670、秋水仙碱、二甲双胍、阿司匹林对迁移体和收缩纤维的形成具有不同程度的影响,迁移体可能在肿瘤、2型糖尿病、炎症、心肌梗死等疾病中发挥重要作用.
Study on effect of different functional compounds on formation of migrasomes
Objective To investigate the effects of 4 different functional compounds,SF1670,colchicine,metformin and aspirin on the formation of migrasomes.Methods The L929 Tspan4-mCherry cell line was constructed by Vigofect chemical transfection method.The cells were divided into control group(complete medium without drug),SF1670 group(4 μmol/L),colchicine group(0.6 μmol/L),metformin group(32μmol/L)and aspirin group(1 μmol/L),and treated for 11 hours,respectively.Live cell imaging experi-ment was used to photograph the migrasomes produced by the cells.Fiji-Image J was used to calculate the number of migrasomes produced and the formation area,diameter and length of retraction fibers.Results Con-focal imaging results showed that the L929 overexpressing Tspan4-mCherry cell line was successfully es-tablished,retraction fibers and migrasomes.Compared to the effect of 4 drugs on the migrasomes,the SF1670 group was decreased(P<0.01),and the formation area and length of shrinking fibers were sig-nificantly inhibited(P<0.000 1);the migrasomes number was decreased in colchicine group(P<0.000 1),and the formation area of shrinking fibers was decreased(P<0.05);and the formation area(P<0.05)and diameter(P<0.000 1)was decreased in metformin group.The aspirin was increased(P<0.01)and the diameter of retraction fibers was slightly shortened(P<0.05).Conclusion SF1670,col-chicine,metformin and aspirin have different effects on the formation of migrasomes and retraction fibers.Migrasomes may play an important role in tumors,type 2 diabetes,inflammation,myocardial infarction and other diseases.
