Effects of naphthoquinone in walnut green husk on antioxidant status and inflammatory factors in psoriasis mice
Objective To study the effects of naphthoquinone in walnut green husk on antioxidant status and inflamma-tory factors in mice with psoriatic inflammatory lesions.Methods The psoriasis model of mice was induced by 42 mg 5%imquimod cream on the back of 60 Balb/c mice for 7 consecutive days.60 successfully modeled mice were ran-domly divided into 5 groups,with 12 mice in each group,namely the model group(coated with 10 mL of physiological saline),the positive drug control group(coated with 42 mg of carbopol ointment),the high-dose group of naphtho-quinone compounds in walnut green skin(coated with 42 mg of 60%fluid extract),the medium dose group(coated with 42 mg of 30%fluid extract),and the low-dose group(coated with 42 mg of 15%fluid extract).Another 12 un-modeled mice were selected as normal group(coated with 10 mL of physiological saline).After 10 consecutive days of intervention,the changes of skin lesions were observed and PASI score was performed;the skin lesions were taken by HE staining,and the histopathological lesion was evaluated using Baker score;the activity of SOD,CAT,GSH-Px and the content of MDA in the back skin tissue of psoriasis mice were detected by spectrophotometry;the contents of TNF-α,IL-17 and IL-23 in serum of psoriatic mice were detected by ELISA.Results After 10 days of intervention with corresponding drugs,the back skin tissue of each group of mice was observed under HE staining microscopy:no abnormalities were found in the skin structure of the normal group of mice;The model group mice showed obvious Munro small abscesses in the epidermal layer,excessive keratinization,reduced granular layer,significantly thickened spinous layer,elongated and undulating spinous processes,and scattered mild inflammatory cell infiltration in the der-mis;The pathological changes in the skin tissue of mice in the positive drug control group and the low,medium,and high dose groups of walnut green skin naphthoquinones were similar to those in the model group,but the excessive and incomplete keratinization of the epidermal layer,reduction of the granular layer,and thickening of the spinous layer were alleviated to varying degrees compared to the model group.Compared with the normal group,the back skin of the model group mice significantly thickened,with erythema and scales appearing,the PASI score increased,Baker score increased,the activities of T-SOD,CAT and GSH-Px in back skin tissue decreased,the content of MDA was increased,and the serum of TNF-α,IL-17 and IL-23 contents were increased,with statistical significance(P<0.05).Compared with the model group,the skin redness,scale area and PASI score were reduced in the positive con-trol group and the walnut naphthoquinone low,medium and high dose groups.The activities of T-SOD,CAT and GSH-Px in positive control group and walnut cephaloquinone high-dose group were increased,the content of MDA was decreased,and the Baker score was decreased.The contents of TNF-α,IL-17 and IL-23 in the positive control group and the medium and high dose groups of naphthoquinones in walnut peel were decreased,the difference was statistically significant(P<0.05).Compared with the positive control group,the PASI score and MDA content were increased in the back skin of mice in low,medium and high dose groups.The activities of T-SOD,GSH-Px and CAT were decreased in low and medium dose groups,while the contents of TNF-α,IL-17 and IL-23 were increased.The contents of IL-17 and IL-23 were increased in high dose group,and the difference was statistically significant(P<0.05).Conclusion Naphthoquinone extract from walnut green husk has better antioxidant and anti-inflammatory effects,and thus improve the symptoms of psoriasis in mice.
