Study on population genetics of mycobacterium tuberculosis in Urumqi city
Objective Population genetic study of mycobacterium tuberculosis in Urumqi to reveal genetic variation,haplotype distribution and population history dynamics.Methods Whole genome sequencing technology was used to sequence mycobacterium tuberculosis populations from seven 7 districts and 1 one county in Urumqi,genotyping by single nucleotide polymorphisms,and carrying out analyses of popula-tion genetic diversity,genetic structure,gene flow,and historical dynamics.Results A total of 457 my-cobacterium tuberculosis population samples were collected,of which 22 haplotypes were detected,with a haplotype diversity(Hd)of 0.890 49 and nucleotide polymorphism(π)of 0.496 83,resulting in a high ge-netic diversity of the population.AMOVA analysis showed that genetic variation in the mycobacterium tu-berculosis population occurred mainly within the population,with a coefficient of genetic differentiation between populations(Fst)of 0.068 9 and a gene flow(Nm)of 3.42,and that there was a high level of gene flow between mycobacterium tuberculosis populations.Neutrality tests and mismatch analyses is sugges-ted that the mycobacterium tuberculosis population in Urumqi may not have experienced a significant rapid expansion event.Conclusion Mycobacterium tuberculosis populations in Urumqi have high heritability and mainly undergo intra-population genetic variation with significant gene flow.
