Effect of Songjie ointment prepared from different substrates on osteoarthritis(OA)rats and joint inflammation
Objective To compare the curative effect of turpentine ointment made of sheep oil and ghee and vaseline on osteoarthritis(OA)rats and the effect of joint inflammation.Methods A total of 80 male SD rats were selected for the study,with 10 serving as blank group.The remaining 70 rats were induced with monoiodoacetate(MIA)into their joints to establish an OA model via joint injection.After successful model establishment,the rats were randomly divided into different groups including a model group(no drugs),a positive control group(0.5 mL/time Xiaozhongzhitong),high-dose Songjie ointment groups using sheep oil/ghee as bases(80 mg/kg),medium-dose Songjie ointment groups(40 mg/kg),low-dose Songjie ointment groups(20 mg/kg),high-dose vaseline group(80 mg/kg),medium-dose vaseline group(40 mg/kg),and low-dose vaseline group(20 mg/kg).Each treatment group received medication 3 times per day for 7 consecutive weeks.After 50 days of drug administration,joint inflammation index scores were compared among each group;levels of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and interferon-y(IFN-y)in serum were measured,white blood cell(WBC)and neutrophil(NEU)counts in serum.Results Compared with control group,the joint index,the serum levels of IL-1β,TNF-α and IFN-y,the Counts of WBC and NEU in the model control group were increased(P<0.05).The joint in-dex,Serum IL-1β,TNF-α,IFN-γ,WBC and Neu count in high,middle and low dose groups of turpentine ointment and high,middle and low dose groups of Vaseline were significantly decreased(P<0.05).Com-pared with middle-dose group,the joint index and the serum levels of IL-1β,TNF-α and IFN-γ in high-dose,middle-dose and low-dose group of vaseline were higher(P<0.05).Conclusion Turpentine paste made of sheep oil and ghee as the base and vaseline as the base can improve joint function,reduce inflam-mation and cartilage destruction,especially turpentine paste made of sheep oil and ghee as the base.
