目的 探讨核糖核酸结合基序蛋白45(RNA-binding motif protein 45,RBM45)在肝癌中的表达及其临床应用价值.方法 应用癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库分析RNA结合蛋白RBM45在肝癌中的表达,并关联临床资料及病理特征进行统计学分析.运用基因富集分析软件GSEA对肝癌组织中与RBM45相关的基因进行KEGG信号通路富集分析.结果 与癌旁组织相比,肝癌组织中RBM45表达明显升高(P<0.001),且RBM45表达水平与患者临床分期、肿瘤分级及预后均显著相关(P<0.05),通过构建单变量和多变量COX风险回归模型分析RBM45表达与临床资料的相关性,结果表明RBM45可做为肝癌患者独立预后的危险因素(HR:2.756 95%CI:1.785-4.255,P=4.813e-06);KEGG富集分析显示,RBM45可能与细胞周期、卵母细胞减数分裂、泛素介导的蛋白水解等信号通路密切相关.结论 RBM45在肝癌组织中高表达,且其高表达的患者预后较差;RBM45表达可作为肝癌患者预后判断的独立标志物.
Expression and clinical significance of the RNA-binding protein RBM45 in primary hepatocellular carcinoma
Objective To investigate the expression RNA-binding motif protein 45(RBM45)in primary hepatocellular carcinoma tissues and its clinical value.Methods The expression of RNA binding protein RBM45 in liver cancer was analyzed by using The Cancer Genome Atlas(TCGA)database,the clinical data and pathological characteristics were analyzed statistically.Gene enrichment analysis software GSEA was used to analyze KEGG signaling pathway enrichment analysis of RBM45 genes in liver cancer tissues.Results Compared with adjacent tissues,RBM45 was highly expressed in liver cancer tissues(P<0.001),and its expression level was closely related to clinical stage(P<0.05),grade(P<0.05)and prognosis.The univariate and multivariate Cox risk regression models were performed for analysis the correlation between RBM45 expression and clinical data.The results showed that RBM45 can be used as a risk factor for the independent prognosis of liver cancer patients(HR:2.756 95%CI:1.785-4.255,P=4.813e-06);The enrichment analysis of KEGG signaling pathways that RBM45 might be closely related to cell cycle,oocyte meiosis and ubiquitin mediated proteolysis.Conclusion RBM45 is highly expressed in liver cancer tissue,and the prognosis of patients with high expression is poor.RBM45 expression can be used as an independent marker for prognosis of liver cancer.
